I began thinking of Prime Numbers yesterday, but abandoned the idea when the "Capital"/"Prime" City idea occured to me!!!

I've been too busy to investigate much today (and it looks like everyone else has, too), but I'm guessing that we're looking for five (hopefully well-known) quintets, each associated in some way (composer's birthplace, where he/she lived or died, or where the piece was first performed) with one of the places.

One of the associations which you mention in brackets is indeed the key to the proverbial door. All 5 quintets, one of them Opus 57, are well-known. The list of cities is in chronological order.

I have a feeling it is more specific than this but - we do have Piano Quintets first performed in these cities by Mozart (Vienna), Schumann (Leipzig), Korngold (Hamburg), Elgar (London) and Shostakovich (Moscow) - hopefully this may inch us forward ..... but then again it may not ...
